Direct buses from Yangon to Ngwe Saung depart in the very early morning around 5am to 6am and the return buses depart around noon. There are many companies offering this trip: Kyal Nan Taw, Asia Dragon, Golden Dragon, JJ Express, G&A Tours, ...
There is also one bus (Kan Htoo Aung Company) departing from the Western Bus Terminal Dagon Ayar in Hlaing Thar Yar Township at 10am, returning in the morning at 7:30am.
The most relaxing way is - for sure - to hire a private taxi from Yangon to Ngwe Saung.

Alternatively you can take one of the frequent buses from the Western Bus Terminal Dagon Ayar to Pathein throughout the day and travel on with a minivan, local bus, tuktuk or car taxi to Ngwe Saung.
Minivans and local buses from different companies from Pathein to Ngwe Saung depart apprximately every 2 hours from around 6:30am in the morning until 3:30pm in the afternoon. For the 55km ride it will take about 1.5 to 2 hours.
The return rides from Ngwe Saung to Pathein depart also from 6:30am in the morning to 3:30pm in the afternoon from the main junction at the southern end of Ngwe Saung town. See the Ngwe Saung town map here.
Please beware that in low season it's likely that some rides are cancelled because of too low demand.
Hiring a car taxi, tuktuk or a motorbike taxi are other options.
